摘要
[方法]越南伯克霍尔德氏菌B418是优良的生物控制剂,在前期研究中其主要应用剂型为可湿性粉剂,但其保存期较短,而微胶囊的应用可解决该问题。利用双流体气流式雾化器制备越南伯克霍尔德氏菌B418的微胶囊,采用单因子实验研究微胶囊制备的合适条件,利用经典加速实验确定微胶囊的保存期。[结果]微胶囊最佳制备条件包括:壁材海藻酸钠20~30 g/L,固化剂氯化钙40~50 g/L,芯材B418菌液1.0×1012 cfu/L,搅拌速度150~200 r/min,保护剂脱脂奶粉500 g/L,乳化剂吐温80 5 g/L,微胶囊释放液为pH值6.8的磷酸缓冲液。在25℃下保存677 d,微胶囊中菌株B418的存活率仍在90%以上。[结论]微胶囊是适合菌株B418的剂型,该剂型的保存期接近2年,完全满足大生产的需求。
[Methods] Burkholderia vietnamiensis strain B418 is a good biocontrol agent.In the previous study,the wettable powder was used for its dosage form with the defect of short retention period.Microencapsulation is a good technology to resolve this problem.The microcapsule of strain B418 was prepared by using double fluid pneumatic atomizer,and the optimized conditions of microcapsule preparation was studied based on single factor test,then the thermal stability was determined based on classic accelerating test.[Results] The optimized conditions of microcapsule preparation were as follows: sodium alginate 20-30 g/L,calcium chloride 40-50 g/L,the cell population of strain B418 with 1.0×1012 cfu/L,mixing speed 150-200 r/min,defatted milk power with 500 g/L,tween 80 with 5 g/L,and the releasing solution was phosphate buffer with pH 6.8.The survival ratio of strain B418 in microcapsule could still maintain over 90% after 677 days stored under 25 °c.[Conclusions] The microcapsule is the suitable dosage form of strain B418 with the retention period of almost 2 years,and it can satisfy the large scale production.
