摘要
目的建立一种由猪血清、四氯化碳、乙醇3种因素刺激的新型肝损伤大鼠模型。方法将56只健康雄性大鼠随机分为A、B、C、D、E 5组,其中A至D组为实验组,E为空白对照组。除空白对照组外,其余各组连续4周多因素诱导肝损伤模型。每周摘眼球取血,测定血清中谷草转氨酶(AST)、谷丙转氨酶(ALT)活性。第4周取血后处死动物,取肝脏进行切片观察。结果多因素诱导肝损伤各模型组AST和ALT均不同程度的升高,与空白对照组相比,在不同时间点上有显著性差异(P<0.05)或非常显著性差异(P<0.01)。病理切片也证实各实验组出现不同程度的肝损伤。结论由猪血清、四氯化碳、乙醇3种因素诱导可建立较为理想的大鼠肝损伤模型。
Objective To establish a novel rat model of liver injury induced by carbon tetrachloride,ethanol,and porcine serum.Methods A total of 56 healthy male rats were randomly divided into 5 groups: A,B,C,D,and E.Group A to D were model groups,while group E was a normal control group.Except for the normal control group,the rats in other 4 groups were fed drugs by complex factors for 4 weeks.The serum levels of AST and ALT were examined every week.At the end of the last week,the liver tissues were obtained from the rats and observed under electron microscopes.Results The serum levels of ALT and AST of group A to D were all significantly different from those in the normal control group(P〈0.05 or P〈0.01).Histopathological section of the liver showed that the liver of the model groups were all injured to different degrees.Conclusion An ideal liver injury model in rats is induced by porcine serum,carbon tetrachloride and alcohol.
