摘要
采用氚标记液闪计数法测定了不同光周期条件下湖北光敏核不育水稻105(简称105s)及其常规品种鄂宜105花粉发育期的 DNA、RNA 及蛋白质的合成动态。并用荧光素-荧光素酶法测定了 ATP 的含量动志。结果表明,长日诱导的105s 的花药在花粉发育过程中核酸及蛋白质的合成活性一直很低,且无明显峰值。其花药 ATP 的含量从花粉发育的早期就明显低于可育花药,随着花粉发育的进程,差异越来越大。由此可知花药中大分子合成水平降低与花粉败育有关。
During the develoment of pollens of HPGMR 105s and cultivar Eyi 105 (Oryza astiva L.subsp.Japonica)induced by short—day(10h/day)and long- day(14h/day),the content of ATP of the anthers was determined by the luci- ferin-luoiferase method,and the protein and nucleic acid synthetic activities in the anthers were investigated by liquid scintillation counter with ~3H-leucine incorperating into protein,~3H-uridine into RNA and ~3H thymidine into DNA.The results show that the protein,RNA and DNA synthetic activities of the anthers of 105s induced by long-day are much lower than those of normal anthers during the pollen development and that there are no peaks of synthesis to appear.The content of ATP in the anthers of 105s induced by long-day is much lower than that in the normal anthers from the early uninucleate stage of pollen grains to mattiration phase.
