摘要
目的 评价盐酸万乃洛韦的细胞毒性和抗单纯疱疹病毒活性。方法 用细胞培养法测定盐酸万乃洛韦和阿昔洛韦对 Ｖｅｒｏ 细胞的细胞毒性和对６ 株单纯疱疹病毒（ 其中Ⅰ型和Ⅱ型各３ 株） 的抗病毒活性。结果 盐酸万乃洛韦对 Ｖｅｒｏ 细胞的半数中毒浓度（ Ｔ Ｄ５０） 为２９２ ．７１μｍ ｏｌ／ Ｌ；对 Ｈ Ｓ Ｖ１ 和 Ｈ Ｓ Ｖ２ 的平均半数有效浓度（ Ｉ Ｃ５０） 分别为０ ．１１１μｍ ｏｌ／ Ｌ 和０ ．１６９μｍ ｏｌ／ Ｌ；对 Ｈ Ｓ Ｖ１ 和 Ｈ Ｓ Ｖ２ 的抗病毒治疗指数分别为２６４０ 和１７３１ 。结论 万乃洛韦是一种细胞毒性小的有效的抗 Ｈ Ｓ Ｖ 药物。
Objective To evaluate the cytotoxicity and antiherpesvirus activity of valaciclovir hydrochloride.Methods The cytotoxicity to Vero cell and antivirus activity to 3 strains of herpes simplex virus type1(HSV 1) and 3 strains of herpes simplex virus type 2(HSV 2) of valaciclovir hydrochloride were detected in vitro.Results The 50% toxicity dose(TD 50 ) to Vero cell was 292.71μmol/L;the average 50% inhibitory concentrations (IC 50 s) to HSV 1 and HSV 2 was 0.111μmol/L and 0.169μmol/L,respectively,the therapeutic indexes for HSV 1 and HSV 2 were 2 640 and 1 731,respectively.Conclusion It is indicated that valaciclovir hydrochloride in an effective anti HSV compound with low cytotoxicity.
