人输卵管Wnt家族部分基因的表达及米非司酮对其表达的影响 被引量:1

The gene expression of Wnt family members in human fallopian tissues and its regulation by mifepristone

摘要 目的了解Wnt基因及其抑制因子在人输卵管上有无表达,并观察其在月经周期中的变化,探讨米非司酮对Wnt基因表达的调节作用。方法选取育龄妇女的正常输卵管组织共18例,分为3组,即增殖期输卵管组织6例、分泌期输卵管组织6例,以及分泌期服用米非司酮50 mg并于24h后手术的输卵管6例。应用实时定量聚合酶联反应(PCR)技术检测各组的Wnt-3、Wnt-5a、Wnt-7a、FrpHE和DKK1的mRNA表达;应用蛋白印迹方法检测FrpHE和DKK1的蛋白表达。结果 Wnt-3、Wnt-5a、Wnt-7a、FrpHE和DKK1在正常育龄妇女的增殖期和分泌期输卵管组织中均有表达。增殖期Wnt-3和FrpHE的mRNA表达要显著高于分泌期(P<0.05),而分泌期DKK1的mRNA表达则显著高于增殖期(P<0.05),其它基因的表达无显著性差异。FrpHE和DKK1的蛋白表达与其mRNA表达相符。分泌期服用米非司酮的输卵管组织的DKK1的mRNA和蛋白表达均较分泌期未服用米非司酮的输卵管组织明显降低(P<0.05),而其余各基因的表达在两组间无显著性差异。结论在正常育龄妇女的输卵管组织中存在Wnt部分家族成员的表达;它们在月经周期中的不同时期发生了相应改变,提示Wnt信号通路可能受到卵巢性激素的调节作用;孕激素可能是DKK1的重要调控因子。 Objective: To investigate the expressions of Wnt family members and their inhibitors in human fallopian tube and the expression changes in different phases of menstrual cycle, and assess the effect of mifepristone on expressions of Wnt genes and their inhibitors. Methods. Fallopian tissue samples were collected from 18 normal fertile women who received salpingooophorectomy or salpingostomy in our hospitals, with 6 in proliferative phase, 6 in secretory phase, and 6 in secretory phase who were pretreated with a signal dose of 50 mg mifepristone for 24 h. Real time PCR technology was usedto detect the expressions of Wnt-3, Wnt-5a, Wnt-7a, DKK1 and FrpHE mRNA in fallopian tubes. The protein expressions of FrpHE and DKK1 were inspected by Western blot. Results. The expressions of Wnt-3, Wnt-Sa, Wnt-7a, FrpHE and DKK1 were all found in fallopian tube in both proliferative and secretory phases. The mRNA expressions of Wnt-3 and FroHE in orolifera tive phase were significantly higher than in secretory phase (P〈0.05), while mRNA expression of DKK1 in secretory phase was significantly higher than in proliferative phase (P〈0. 05). No significantly difference was found in the expressions of the other genes between phases. The protein expression of FrpHE and DKK1 showed the same pattern as mRNA expression. The protein and mRNA expressions of DKK1 were significantly lower in mifepristone-treated secretory phase than those in normal secretory phase (P〈 0.05), while the other genes showed no significant difference in the expression levels between these two groups. Conclusions: Expressions of Wnt-3, Wnt-5a, Wnt-Ta and their inhibitors FrpHE and DKK1 exist in women's fallopian tube. The expression changes during different phases of menstrual cycle implies that Wnt signaling pathway is subject to the regulation of steroid hormones produced by ovary. Progesterone might be an important regulator of the expression Of DKK1.
