胰腺癌MMP-2和TIMP-2表达与预后的关系 被引量:8

Prognostic values of matrix metalloproteinase 2 and tissue inhibitor of metalloproteinase 2 expression in pancreatic cancer

摘要 目的:探讨胰腺癌组织基质质属蛋白酶2( MMP2) 和基质金属蛋白酶抑制剂2(TIMP2) 表达与肿瘤侵袭转移和预后的关系。方法: 通过免疫组化方法检测35 例胰腺癌病理标本和10 正常胰腺标本MMP2 和TIMP2 的半定量表达情况及其与肿瘤临床病理参数和预后的关系,生存分析采用Kaplanmeier 方法。结果:胰腺癌MMP2 和TIMP2 表达阳性率明显高于正常对照组, 与淋巴结转移和临床分级密切相关, 但与胰腺癌的分化程度无关,Kaplanmeier 分析发现MMP2 和TIMP2 高表达患者的生存时间明显低于MMP2 和TIMP2 低表达患者( P<0-001) 。结论:胰腺癌MMP2 和TIMP2 表达与肿瘤侵袭转移和预后密切相关,有助于确定术后治疗方案。 Objective:To investigate the roles of matrix metalloproteinase 2 (MMP 2) and tissue inhibitor of metalloproteinase 2(TIMP 2) in tumor invasion and metastasis or as a prognostic factor in patients with pancreatic cancer. Methods:Tissues were obtained from 35 patients with pancreatic cancer and 10 patients without pancreatic disease. Expression of MMP 2 and TIMP 2 was examined by immunohistochemical technique and scored semiquantitatively (score 0 3) in carcinoma cells, and the results were correlated with clinicopathological tumor parameters.Survival analyses were done using the Kaplan Meire method. Results:Expression of MMP 2 and TIMP 2 was significantly higher in patients with pancreatic cancer than that in normal persons. Expression of MMP 2 and TIMP 2 was significantly correlated with lymph node metastasis and TNM clinical staging sysfem,and did not correlate with histologic classification.Kaplan Meire analysis revealed an association of increasing MMP 2 and TIMP 2 expression with worse prognosis (P< 0 001). Conclusion:MMP 2 and TIMP 2 are believed to contribute to the invasive properties of pancreatic cancer,which can be used to evaluate the prognosis of patients and may be helpful in designing treatment protocols.
