摘要
目的:观察通冠丸对动脉粥样硬化大鼠血清C反应蛋白(CRP)及白介素-6(IL-6)含量的影响,从炎症方面探讨通冠丸延缓动脉粥样硬化形成、发展的可能机制。方法:将健康SPF级Wistar雄性大鼠50只采用随机数字表法分为空白组、模型组、阿托伐他汀钙组、通冠丸组、通冠丸+阿托伐他汀钙组,每组10只。空白组大鼠给予普通饲料喂养,其他各组大鼠均给予高脂饲料喂养,并于实验初始给予维生素D3注射液60万U/kg腹腔注射,分3天完成,以复制动脉粥样硬化大鼠模型,从实验第5周开始灌胃,灌胃8周。于实验第12周末,将各组大鼠禁食12小时,晨起空腹,给予各组大鼠25%乌拉坦(0.5 mL/100 g)腹腔注射麻醉,股动脉采血,提取血清,待测指标,处死动物,取主动脉,进行苏木素-伊红(HE)染色,光镜下观察病理形态学改变。结果:阿托伐他汀钙组、通冠丸组、通冠丸+阿托伐他汀钙组IL-6、CRP水平较模型组减低(P<0.05),通冠丸组与阿托伐他汀钙组无显著性差异(P>0.05),通冠丸+阿托伐他汀钙组水平减低更显著(P<0.01);HE染色法结果显示阿托伐他汀钙组、通冠丸组、通冠丸+阿托伐他汀钙组较模型组大鼠动脉粥样硬化病变减轻,且阿托伐他汀钙+通冠丸组减轻最明显。结论:通冠丸能够降低动脉粥样硬化大鼠血清C反应蛋白及IL-6的水平,延缓动脉粥样硬化的形成、发展。
Objective:To observe the effect of TongGuanWan on serum c-reactive protein and IL-6 levels of experiment atherosclerosis rats,and to study TongGuanWan delaying possible mechanism of atherosclerotic formation and development.Methods:50 male health wistar rats were divided randomly into blank group,model group,atorvastatin group,TongGuanWan group,TongGuanWan combined with atorvastatin group.The group of blank rats were given ordinary feed,the other groups were given high fat feed,and to give vitamin D3 injection 60 million U/kg at the beginning,on the fifth week filling stomachs started,once a day,at 12 weekends,each rat was given 25% urethane intraperitoneal injection of anesthesia,and their femoral artery blood were taken to detecte IL-6 and CRP.meanwhile,the pathomorphoiogy of main artery was performed.Results:(1)Compared with the group of model,the level of serum IL-6 and CRP decreased significantly in the group of TongGuanWan combined with atorvastatin group(P0.01),and were remarkably lower than in the group of atorvastatin(P0.05) and the group of TongGuanWan(P0.05).However,there was no significant difference between the group of atorvastatin and the group of TongGuanWan(P0.05).(2)The pathomorphoiogy showed that the intima of model group apparently were thicker than TongGuanWan combined with atorvastatin group,atorvastatin group or TongGuanWan group,TongGuan pill combined with atorvastatin group was thinner than atorvastatin group or TongGuanWan group,there was no significant between atorvastatin group and TongGuanWan group.Conclusion:(1) TongGuanWan could reduce serum c-reactive protein and IL-6 levels of experiment atherosclerosis rats and delay atherosclerotic formation and development.
