拉米夫定与甘草次酸合用的大鼠药动学研究 被引量:1

摘要 研究大鼠灌胃拉米夫定(lamivudine,简称3TC,25 mg/kg)联合甘草次酸(glycyrrhetic acid,简称GA,50 mg/kg)前后的药动学变化.Wistar大鼠分别单次灌胃3TC,GA以及3TC+GA(25 mg/kg+50 mg/kg)后,于不同时间点采集大鼠血浆,血浆样品经处理后,采用HPLC法测定各时间点血浆中3TC和GA的含量.血药浓度-时间数据用Win Nonlin5.2.1药动学软件计算药动学参数,采用SPSS statistics17.0软件对合用前后参数进行统计分析.合用后,3TC的Tmax和t1/2(ka)显著变大、GA的Cmax和AUC0~48 h均显著变大,说明3TC与GA联合使用,显著影响了3TC与GA的吸收,对其他药动学参数无显著影响. It is the object to study pharmacokinetics of combined therapy of lamivudine(3TC,25 mg/kg) and glycyrrhetic(GA,50 mg/kg) acid in rats.Wistar rats were orally administered a single dose of 3TC,GA and 3TC + GA(25 mg/kg+50 mg/kg) respectively.The plasma samples were collected at different time after oral administration.The contents of 3TC and GA in plasma were determined by HPLC method established in the present work.The concentration-time data of 3TC and GA were processed by WinNonlin5.2.1 pharmacokinetic software.The pharmacokinetic parameters were analyzed statistically by SPSS statistics17.0 software.The results indicated that compared with the pharmacokinetic parameters of 3TC or GA sole administration group,Cmax and AUC0-48 h of GA,t1/2(ka) and Tmax of 3TC were significantly increased in the co-administration group of 3TC + GA.Based on the results,the co-administration of 3TC + GA has an important influence on the absorption process,no obvious influence on the distribution,metabolism and excretion process of 3TC and GA in rats.
