摘要
目的:探究豆梨多糖提取的技术方法和最佳工艺条件。方法:样品采用恒温水浴浸提法提取,用醇沉法得到多糖沉淀,用稀硫酸溶解后,采用苯酚-硫酸法,在波长490nm处测定吸光度,以葡萄糖标准品为对照品,计算豆梨多糖的提取率;采用正交实验法,对影响多糖提取的料液比、提取时间、提取温度等3个因素进行考察,确定豆梨多糖提取的最佳工艺条件,并对结果进行验证。结果:最佳提取工艺条件为:料液比1∶15、浸提温度95℃、浸提时间2h。此条件下得到的多糖提取率最高,平均产率为3.04%。
Objective:To explore the technical methods and optimum conditions of polysaccharide from Pyrus calleryana Decne. Methods:Samples were extracted by constant temperature water extraction,polysaccharide precipitation was made by alcohol precipitation method. After dissolving with dilute sulfuric acid,the polysaccharide extracted from P. calleryana was determined with phenol-sulfuric acid method,the absorbance was determined at the wavelength of 490 nm,glucose was choose as reference standard to calculate the extraction rate of P. calleryana polysaccharide. Use of the orthogonal experiment method,three factors impacted the extraction rate of polysaccharide,such as the extraction liquid ratio,extraction time,extraction temperature,were studied. The optimum conditions of polysaccharide extraction of P. calleryana Decne was determined,and the results are verified. Results:The optimum extraction conditions are as follow:liquid ratio is 1∶15,extraction temperature is 95 ℃,extraction time is 2 h. Under these conditions,the extraction rate of polysaccharides extracted from P. calleryana Decne was the highest. The average production rate was 3.04%.
