摘要
以根结线虫天敌1号分离物为提取碱性磷酸酶的材料,菌体经液氮研磨破碎后,用正丁醇抽提,硫酸铵分级沉淀分离,透析获得酶的粗提取液。再经过DEAE-52离子交换和SephadexG-150分子筛两步层析得到纯化倍数为27.17、比活力达10 514U/mg的碱性磷酸酶。
The study aimed to isolate the alkaline phosphatase from destroying fungi of root-knot nematode-1（DFRKN-1）.It had been isolated by means of the following procedures： n-butanol extraction and ammonium sufate fractionation,the partially purified enzyme was gained by means of ion-exchange chromatography on DEAE-52,and gel filtration chromatography on Sephadex G-150.The purification attained to 27.17 folds and the specific activity was 10 514 U/mg.
