禁食对大鼠肝脏胰岛素样生长因子-Ⅰ及其结合蛋白-lmRNA表达的影响 被引量:5

摘要 目的探讨营养状态调节机体生长发育的分子生物学机制。方法利用Northern印迹技术,分析禁食对雄性Wistar大鼠肝胰岛素样生长因子-Ⅰ(IGF-Ⅰ)和胰岛素样生长因子结合蛋白(IGFBP-Ⅰ)基因表达的影响。结果禁食48小时后大鼠体重下降11.8%、血糖下降50.8%;禁食48小时后,肝IGF-ⅠmRNA含量下降32.7%;与此相反,禁食后肝IGBP-1mRNA含量逐渐上升,禁食24小时后IGFBR-ⅠmRNA含量为正常的2.5倍;禁食后肝白蛋白mRNA含量无明显变化。结论禁食抑制大鼠肝IGF-Ⅰ基因表达,促进IGFBF-Ⅰ基因表达。 Objective To investigate the molecular mechanisms of body development and growth regulatedby nutritional status.Methods Effects of fasting on the expression of insulin-like growth factor-Ⅰ (IGF-Ⅰ)andIGF binding protein-1 (IGFBP-1)genes were analyzed by northern blotting technique in rats.Results Fasting for48 hours, body weight of the rats decreased by 11.8%,level of blood glucose by 50.8% and content of IGF-ⅠmRNA in the liver by 32.7%(P<0.05).On the contrary, content of IGFBP-1 mRNA in the liver of fasted ratsincreased gradually, with a 2.5 time increase after 24 h fasting (P<0.05). There was no sigificant change inthe level of liver albumin mRNA after fasting. Conclusion Fasting could inhibit the expression of IGF-Ⅰ geneand enhance the expression of IGFBP-1 gene.
