摘要
目的:探讨补阳还五汤对异种去细胞神经支架移植后神经再生及功能恢复的影响以及与GM1(mono-sialotetrahexosy 1 gangliosides)的效果比较。方法:36只健康成年SD大鼠随机分为3组:补阳还五汤(BYHWD)治疗组、GM1治疗组和生理盐水(NS)对照组。动物分别存活4、8周,不同时段各组为6只。以上3组大鼠分别以10 mm长度的兔异种去细胞神经支架桥接大鼠坐骨神经缺损。术后补阳还五汤组和生理盐水对照组分别用补阳还五汤水煎剂和生理盐水按10 ml/kg灌胃,持续1个月。GM1治疗组术后大鼠损伤侧小腿三头肌内注射GM1 0.1 ml/d;各组分别在术后第4周、8周行大体观察、测定坐骨神经运动传导速度恢复率(MNCV%)和小腿腓肠肌复合动作电位波幅恢复率(CMAP%)、移植体的神经纤维行电镜半薄切片观察记数有髓纤维的数目,免疫组织化学标记NF和S-100。结果:在同一时相点补阳还五汤组坐骨神经运动传导速度恢复率、小腿腓肠肌复合动作电位波幅恢复率(CMAP%)及移植体的有髓纤维数目均优于生理盐水对照组(P<0.05)。补阳还五汤组与GM1组比较没有显著性差异(P>0.05)。结论:应用补阳还五汤可促进异种去细胞神经支架移植后神经再生与功能恢复;补阳还五汤和GM1对异种神经支架移植后神经再生与功能恢复的效果没有显著性差异。
Objective:To explore the effects of Buyang Huanwu decotion(BYHWD) on nerve regeneration and functional recovery after xenogeneic acellular nerve scaffold grafting and compare the effects of BYHWD with monosialotetrahexosy 1 gangliosides(GM1).Methods:Thirty-six healthy Sprague-Dawley rats were randomly divided into 3 groups:BYHWD-treated group,GM1-treated group and normal saline control group.The rats survived for 4 weeks or 8 weeks(n=6).Sciatic nerve defects of rats were bridged with 10 mm long rabbit xenogeneic acellular nerve scaffold.After the operation,the BYHW decotion treated group and normal saline control group were fed on BYHW decotion or normal saline at the dosage of 10 ml/kg for 30 days.The GM1-treated group was injected with 0.1 ml of GM1 on the surgery leg once a day for two weeks.At 4 weeks and 8 weeks after the operation,a series of measurement were performed including recovery rate of the motor nerve conduction velocities(MNCV%),hind limbs recovery rate of the complex muscular action potential(CMAP%),numbers of myelinated nerve fibers in grafting part,immunohistochemical staining of nerve fine NF and S-100 in grafting part.Results:At the same time span,the recovery rate of the MNCV,hind limbs recovery rate of the CMAP,and numbers of myelinated nerve fibers in grafting part in the BYHWD treated group were better than those in normal saline-treated group(P0.05).However,there was no significant difference between BYHWD group and GM1 group(P0.05).Conclusion:BYHWD can enhance peripheral nerve regeneration and functional recovery in xenogeneic acellular nerve scaffold grafting,but there was no difference on the above mentioned indexes between the two sole-treated groups.
