摘要
目的探讨中药壁虎对食管癌大鼠的治疗作用及其对食管组织环氧化酶-2(COX-2)mRNA转录水平的影响。方法甲基苄基亚硝胺皮下注射建立Wistar大鼠食管癌模型,分为模型组、中药组,化疗组及中药联合化疗组,分别以生理盐水、中药壁虎水煎液、替加氟、中药壁虎水煎液联合替加氟灌胃给药,并以同期正常饲养大鼠为正常对照组。连续给药30 d后,取食管组织进行病理形态学及病理组织学分析,提取食管组织总RNA,RT-PCR法检测COX-2 mRNA转录水平。结果替加氟可明确改善食管癌大鼠的食管组织病理形态,壁虎水煎液对于食管癌大鼠的病理形态无明显改善作用;替加氟及壁虎水煎液均可在数值上降低食管癌变率,但差异无统计学意义;壁虎水煎液可下调食管组织COX-2 mRNA的转录水平。结论下调食管组织COX-2 mRNA转录水平是中药壁虎防治食管癌的可能机制之一,但中药壁虎在食管癌防治方面的疗效尚缺乏大样本临床证据支持,因此,壁虎在防治食管癌领域是否具有开发应用价值有待商榷。
Objective To investigate the effect of gecko on the transcription level of COX-2 mRNA in esophageal carcinoma model rats.Methods Rat esophageal carcinoma model was established by being injected with MBNA.Forty model rats were randomly divided into model group,gecko decoction group,tegafur group and combined group.Thirty days later the rats were killed and the general situation and the pathological changes of esophageal mucosa were observed.We also detect the transcription lever of COX-2 mRNA by RT-PCR method.Results Tegafur could clearly improve pathologic morphological situation in esophageal cancer rats while gecko decoction could not.Tegafur and gecko decoction could depress in value of esophageal cancer rates,but the difference was not statistically significant.Gecko decoction can lower COX-2 mRNA transcription levels in esophageal tissues.Conclusion It is suggested that lowering COX-2 mRNA transcription levels may be one of possible mechanisms for gecko in prevention and treatment of esophageal cancer,but a large sample of clinical evidence is still needed for supporting the efficacy of gecko in cancer prevention.Therefore,the value of gecko in the fight against esophageal cancer is still open to question.
