三种大黄酚制剂改善脑缺血/再灌注小鼠记忆功能的实验筛选研究 被引量:18

摘要 目的研究大黄酚聚氰基丙烯酸丁酯纳米囊、大黄酚羟丙基-β-环糊精包合物、大黄酚脂质体对脑缺血/再灌注(I/R)小鼠记忆功能的保护作用,以期选出药效最佳的大黄酚制剂。方法采用小鼠脑I/R损伤模型,进行避暗、探索、跳台、穿梭实验,观察3种大黄酚制剂对小鼠记忆功能的保护作用;并对各剂量组血液中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活力进行测定,以及小鼠断头缺氧后存活时间和脑指数的测定。结果 3种大黄酚制剂可明显改善脑I/R损伤所致的记忆障碍;提高SOD、GSH-Px活力,延长缺氧后存活时间和增加脑指数,以大黄酚脂质体的作用最为明显。结论 3种大黄酚制剂对脑I/R损伤小鼠记忆功能具有保护作用,以大黄酚脂质体的作用最佳。 Aim To study the protective effects of chrysophanol loaded polybutylcyanoacrylate nanocapsules,chrysophanol-hydroxypropyl-β-cyclodextrin Inclusion Complex,Chrysophanol liposomes on learing and memory impairment in mice with cerebral ischemia reperfusion,and to select the best efficacy of chrysophanol preparation.Methods Cerebral ischemia reperfusion injury was produced on conscious mice by temporarily obstructing bilateral common carotid arteries.The protective effects of three chrysophanol formulations on learing and memory impairment in mice were observed using passive avoidance test,exploratory movement test,platform test and shuttle test,and the glutathione peroxidase(GSH-Px)activities in blood,the superoxide dismutase(SOD)activities in plasma,survival time in beheaded hypoxia mice and brain index were measured.Results Three chrysophanol formulations could improve the impairments of memory with cerebral ischemia reperfusion,and increase the glutathione peroxidase(GSH-Px)activities in blood and the superoxide dismutase(SOD)activities in plasma,and prolong the survival time in beheaded hypoxia mice,and increase the brain index.The most significant role of it was chrysophanol liposomes.Conclusion Three chrysophanol formulations show protective effects on learning and memory of mice with cerebral ischemia reperfusion,and chrysophanol liposomes is the most significant.
