摘要
目的:测定苦荞提取物中芦丁和槲皮素的含量,评估苦荞提取物质量。方法:Welchrom-C18色谱柱(4.6 mm×150mm,5μm),流动相甲醇-0.2%甲酸水溶液(50∶50),检测波长280 nm,柱温30℃,流速1.0 mL·min-1,进样量10μL。结果:芦丁在1.56～100.00 mg·L-1,槲皮素在1.52～97.00 mg·L-1呈良好的线性关系。平均回收率芦丁为98.49%(RSD 1.25%,n=5),槲皮素为97.87%(RSD 1.69%,n=5),芦丁的含量为18.97%±1.43%,槲皮素的含量为5.03%±0.42%。结论:该方法成功应用于苦荞提取物中的芦丁和槲皮素的含量测定。
Objective：To determine the content of rutin and quercetin in the tartary buckwheat extract,so as to appraise its quality.Method： The chromatographic conditions were as follows： with the Welchrom-C18（4.6 mm × 150 mm,5 μm） as the chromatographic column,methanol-0.2% formic acid solution（50∶ 50） as the mobile phase,280 nm as the detection wavelength,30 ℃ as the column temperature,1.0 mL·min-1 as the flow rate.Result： The linear range for rutin was 1.56-100.00 mg·L-1 and the linear range for quercetin was 1.52-97.00 mg·L-1.The average recovery for rutin and quercetin was 98.49%（RSD 1.25%,n = 5） and 97.87%（RSD 1.69%,n = 5） respectively.Conclusion： Rutin and quercetin were 18.97% ± 1.43% and 5.03% ± 0.42% respectively determined by the method applied.
