摘要 目的探讨骨髓间充质干细胞(MSCS)在雨蛙素联合脂多糖导致的重症急性胰腺炎中的作用。方法采用雨蛙素联合脂多糖造成大鼠重症急性胰腺炎模型,经静脉移植体外扩增培养并用Hoechst33258标记的异体大鼠MSCS,检测损伤后0.5,2,4,6,12 h大鼠血浆中的TNF-α,IL-6的水平变化。以及不同时间段胰腺和肺组织病理变化。同时观察标记的MSCS在胰腺,肺脏和肝脏中的分布情况。结果与对照组相比,MSCS能降低治疗组大鼠血浆中各个时间点的TNF-α,IL-6的水平,同时降低肺湿/干重比值,减轻胰腺和肺脏炎性损伤,标记的MSCS在胰腺,肺脏和肝脏中的分布有明显差异。结论 MSCS移植对大鼠重症急性胰腺炎具有保护作用。 Purpose To investigate the role of bone marrow mesenchymal cells in severe acute pancreatitis(SAP) in rat.Methods SAP model was induced by receiving intraperitoneal injection with caerulein and lipopolysaccharide(LPS).Rat MSCS were isolated and cultured in vitro,stained with Hoechst33258 and transplanted in rats.The level of plasma TNF-a and IL-6 was detected at 0.5,2,4,6,12 h after caerulein and LPS injection.The pathological changes of pancreas and lung,and the distribution of stained MSCS in pancreas lung and liver were detected at the same time.Results In comparison with control group,the plasma cytokine of TNF-α,IL-6 expression was suppressed,and lung wet dry ratio lowered at different time points after injection of MSCS.The injury of pancreas and lung was reduced.The difference of distribution of stained MSCS between these organs is significant.Conclusion MSCS have significantly protective effects in SAP of rat.
