摘要
目的体外培养人脐静脉血管内皮细胞(HUVEC),加入低密度脂蛋白(LDL)建立细胞损伤模型,观察脂联素(APN)对模型细胞结构和功能的影响。方法观察对照组、LDL损伤组、APN预处理组和APN干预组内皮细胞(EC)的形态改变,测定细胞活性,测定细胞培养上清中一氧化氮(NO)、乳酸脱氢酶(LDH)和6-酮前列腺(6-keto-PGF1)的含量变化,测定细胞中一氧化氮合酶(NOS)的含量。结果与正常细胞对照组比较,LDL损伤组的细胞形态明显改变,细胞存活率明显降低,LDH释放明显增高,NO、NOS和6-keto-PGF1合成明显降低(P<0.01);而APN预处理组和干预组能明显降低LDH释放,促进NO、NOS和6-keto-PGF1合成(P<0.01)。结论 APN对EC有一定的保护作用,能拮抗LDL对EC的损伤。
Objective To observe the effect of lipoprotein (LDL) on the cultured human umbilical vein endothelial cells (ECs) damage models established by adding low-density lipoprotein (LDL) in vitro and to observe the effect of adiponectin (APN) on the Ecs.Methods The cultured ECs were divided into five groups. GroupⅠ:normal Ecs; groupⅡ:ECs damaged by LDL;group Ⅲ:ECs cocultured with APN;group Ⅳ:LDL injuried model pre-treated with APN;group Ⅴ:LDL injuried model treated with APN. The morphological changes and survival rate of the ECs were observed,and the levels of nitric oxide (NO),nitric oxide synthase (NOS),lactate dehydrogenase (LDH) and 6-keto prostaglandin (6-keto-PGF1 ) were detected.Results As compared with the normal ECs,the cell morphous in LDL injury group were significantly changed,and the survival rate was significantly lower,and the levels of LDH were significantly increased,however,the levels of 6-keto-PGF1,NO and NOS were significantly decreased(P〈0.01).When the ECs pre-treated or treated with APN,the release of LDH was inhibited,and the synthesis of 6-keto-PGF1,NO and NOS was obviously enhanced(P〈0.01).Conclusion The APN has certain protective effect,which can relieve the damage of LDL on EC.
