摘要
目的 通过检测大鼠随意皮瓣中ET和TXB2的含量,探讨局部应用天然及重组的水蛭素对随意皮瓣静脉淤血的影响.方法 自2009年4月至2009年8月,选取健康的Wistrar大鼠21只,随机分为A、B、C 3个组,于所有大鼠的背部设计并制作超长随意皮瓣淤血模型后,A组局部注射天然水蛭素;B组局部注射重组水蛭素;对照C组局部注射生理盐水.于皮瓣远端切取组织块,检测内皮素(ET)、血栓素B2(TXB2)的含量.计算各组皮瓣的成活率,取成活皮瓣组织行常规HE染色后计数炎性细胞和微血管的数量.结果 各组中ET-1的含量:注射后第5天和第7天,C组中ET-1的含量高于A、B两组,其差异有统计学意义(P〈0.05).各组中TXB2的含量:注射后第3天和第5天,C组中TKB2的含量高于A、B两组,其差异有统计学意义(P〈0.05).A组中TXB2的含量高于B组,但两组比较其差异无统计学意义(P〉0.05).结论 局部应用天然及重组的水蛭素使移植后的大鼠随意皮瓣中ET、TXB2的含量降低.由此推断,天然及重组的水蛭素对移植后随意皮瓣的静脉淤血可有改善作用.
Objective To explore the effect of natural and recombinant hirudin on vein congestion of rat random flap by detecting the content of ET, TXB2 of the flap. Methods From April 2009 to August 2009, 21 heath Wistrar Rats were selected and divided randomly into three groups (Group A, B, C). Over-long random flap models with congestion were designed and established on the back of the Rats. The natural hirudin, the recombinant hirudin and saline were injected locally in Group A, Group B and Group C (the control group) respectively. The context of endothelin (ET), thromboxane B2 (TXB2) in tissue blocks taken from some distal flap were detected. The survival rate of the flaps in each group were calculated and the amounts of the number of inflammatory cell and capillary vessel were observed in survival flap after HE staining. Results The content of HE-1 in Group C was higher than that Group A and B at 5, 7 days after injection. Its difference had statistical significance ( P 〈 0.05 ). The content of TXB2 in Group C was higher than that in Group A and B at 3, 5 days after injection. Its difference had statistical significance ( P 〈 0.05), which in Group A was also higher than that in Group B, but there was no statistical significance between them ( P 〈 0.05). Conclusion The ET, TXB2 levels in flag were decreased by injecting locally the natural and recombinant hirudin, thereby we can conclude that natural and recombinant hirudin on random flap transplantation may play a role of improving venous congestion
