摘要
目的:探索3日龄幼鼠单侧颈总动脉结扎联合缺氧造成脑白质损伤后少突胶质细胞及髓鞘超微结构的变化。方法:将3日龄未成熟SD新生大鼠随机分为对照组和实验组。实验组大鼠结扎右侧颈总动脉,置8%O2和92%N2混合气体的缺氧装置2h后,取出返笼饲养。对照组仅游离右侧颈总动脉,并未结扎及缺氧实验。术后28d透射电镜观察胼胝体少突胶质细胞及髓鞘超微结构。结果:对照组大鼠少突胶质细胞染色质分布均匀,细胞器结构完整;髓鞘厚度及数目均正常;实验组大鼠部分少突胶质细胞核固缩、核破碎,细胞器广泛缺失;与对照组相比髓鞘厚度变薄,数目明显减少。结论:3日龄幼鼠脑缺血缺氧28d后,脑白质少突胶质细胞的超微结构发生了坏死性病理改变,髓鞘形成能力减弱,髓鞘化轴突的数目减少。
Objective:To investigate the changes of ultrastructures of oligodendrocyte and myelin sheath following hypoxic-ischemic injury in 3-day-old neonatal rats.Methods:3 day-old Sprague-Dawley(SD)premature rats were randomly divided into experimental and control groups.Hypoxic-ischemic brain injury of the experimental group was induced by ligation of right carotid artery and then in the 8%O2/92%N2 hypoxia installation 2 hours.The control group underwent a sham operation without carotid ligation and hypoxia.At 28 days after operation,the oligodendrocytes and myelin sheath of corpus callosum were observed by transmission electron microscopy.Results:The chromatin well-distributed and organelles structures integrity in oligodendrocytes,and thickness and number of myelin sheath were normal in the corpus callosum in control group rats.In the experimental rats,oligodendrocytes karyopyknosis and disintegration and organelles absence widespreadly were observed.The myelin sheath thickness became thin and the number of myelin sheath decreased compared with control rats.Conclusion:Necrotic pathological changes of ultrastructures of oligodendrocytes and the decreased number of myelinated axon were induced by hypoxic-ischemic brain injury in 3-day-old neonatal rats.
