摘要
目的:观察葛根素对人食管癌细胞EC9706生长的影响。方法:不同浓度的葛根素作用于人食管癌细胞EC9706,采用四甲基偶氮唑蓝(MTT)法检测药物对细胞的增殖抑制率;流式细胞术(FCM)检测细胞周期分布。透射电镜检测葛根素注射液处理的EC9706细胞形态学表现。结果:葛根素能明显抑制食管癌细胞EC9706的增殖,并呈剂量和时间依赖性,其半抑制率(IC50)为20.1mg/l;经IC50葛根素处理食管癌细胞EC9706后,FCM分析发现实验组G0/G1期EC9706细胞与对照组相比均有显著增多,G2/M期细胞逐渐减少;透射电镜发现葛根素处理的EC9706细胞呈凋亡的形态学表现。结论:葛根素能明显抑制人食管癌细胞EC9706的增殖,诱导其凋亡。
Objective:To observe the growth influence on human esophageal cancer cells EC9706 induced by puerarin.Methods:EC9706 cells were treated with puerarin of the different concentrations.MTT method was used to detect the growth inhibition effects.Flow cytometry was used to detect the proliferation of the cells after staining by PI or Annexin V-FITC and PI.Electron microscope was used to study the morphological character of EC9706 treated by puerarin.Results:The EC9706 cell growth was inhibited significantly,with a time and dose dependent manner.After puerarin treatment,the apoptosis of EC9706 cells were induced.FCM analysis found that the number of treated cells in G0/G1 phase increased,but that in G2/M phase decreased.Morphological apoptosis of EC9706 cells could be seen under electron microscope.Conclusion:Puerarin can significantly inhibit proliferation and induce apoptosis of human esophageal cancer EC9706 cells.
