动态增强磁共振成像评估血管升压素缺乏鼠肾功能及其对血管升压素治疗的反应

Evaluation of Kidney Function and Response to Vasopressin in Vasopressin-Deficient Rats Treated by Arginine Vasopressin by Using Dynamic Gadolinium Diethylenetetramine Pentaacetic Acid Enhanced Magnetic Resonance Imaging

摘要 目的使用动态增强钆喷替酸葡甲胺(Gd-DTPA)磁共振(MRI)评估先天性血管升压素(AVP)缺乏鼠(Brattleboro鼠,BB鼠)肾脏浓缩稀释功能及BB鼠肾脏对AVP长期或快速治疗的反应。方法实验动物为3周龄雄性BB鼠21只和正常雄性慕尼黑鼠(Wistar鼠)7只。7只BB鼠长期注射AVP(6 d),7只BB鼠快速注射AVP(2 h),另7只BB鼠不接受AVP治疗(BB鼠对照组),7只正常Wistar鼠作为Wistar鼠对照组。在注射Gd-DTPA的0~60 min采用动态增强MRI检查,并记录其肾脏皮质、髓质和肾盂的相对信号强度(RSI)和肾脏血流量(RBF),采用SPSS11.0软件分别分析每组数据。结果与BB鼠对照组比较,BB鼠肾脏RSI曲线发生明显改变。长期和快速注射AVP治疗组与Wistar鼠对照组比较RSI曲线均无明显差别。BB鼠双侧RBF值较Wistar鼠高。快速注射和长期注射AVP均能使RBF增多。结论 Gd-DTPA动态增强MRI可以为检测肾脏AVP改变和评估肾脏功能提供重要信息。无论是快速注射AVP还是长期注射AVP,动态增强MRI均为检测肾内AVP浓度的一种灵敏方法。 Objective To evaluate the kidney concentration and dilution function and response to arginine vasopressin(AVP) as a chronic infusion or as an acute injection in vasopressin-deficient rats(Brattleboro rats,BB rats)by using dynamic gadolinium diethylenetetramine pentaacetic acid(Gd-DTPA)enhanced magnetic resonance imaging(MRI).Methods The study included 21 male 3-week-old BB rats and 7 male 3-week-old Wistar rats.Treatment of AVP was given in 14 BB rats either as a chronic infusion(6 days,n=7)or as an acute injection(2 hours,n=7).Other 7 BB rats were not been treated with AVP(BB rats control group).Seven Wistar rats were used as normal Wis-tar rats control group.Dynamic MRI was performed at 0-60 minutes following the administration of Gd-DTPA(0.05 mmol·kg-1)and the mean relative signal intensity(RSI)and renal blood flow(RBF) were measured.Data of each group were analyzed separately by SPSS 11.0 software.Results A different renal RSI patterns were found between vasopressin-deficient kidneys of BB rats and those of BB rats control group.Supplement of vasopressin in BB rats resulted in patterns resembling RSI curves of Wistar rats control group.The RBF of BB rats was higher than that of Wistar rats and increased after treatment of AVP either as a chronic infusion or as an acute injection.Conclusions These results demonstrate that dynamic MRI may provide useful information for distinguishing between kidneys having changes in the renal vasopressin concentration and renal function.Either as a chronic infusion or as an acute injection,dynamic Gd-DTPA enhanced MRI is an acute methods of detecting AVP concentration in kidney.
