HSL表达和活性与豚鼠血清睾酮水平的相关性

Correlation between the Activity and Expression of HSL and the Serum Testosterone Level in Guinea Pigs

摘要 目的研究不同发育阶段的豚鼠睾丸和附睾组织中激素敏感性脂肪(hormone-sensitive lipase,HSL)的表达及活性与生殖能力成熟的关系。方法取不同天龄阶段的豚鼠18只分为3组:6d龄幼鼠组;16d龄青春期豚鼠组;70d龄成年豚鼠组。用RT-PCR方法检测睾丸和附睾组织中HSL的mRNA表达,37℃下比色法对酶活性进行测定,放免法测定血清中睾酮水平,HE染色睾丸和附睾组织切片进行组织形态学分析。结果发育阶段的豚鼠随着天龄的增加,睾丸特异性HSL mRNA表达也增加(P<0.05),酶活性增强(P<0.05),血清中睾酮水平升高(P<0.01)。组织病理学显示16d的豚鼠睾丸和附睾中开始出现精子,70d精子发育成熟。HSL的表达和酶活性与血清睾酮水平高度相关(r=0.75,P<0.01;r=0.96,P<0.01)。结论发育成熟豚鼠睾丸和附睾组织有较强的HSL表达及活性,HSL与睾丸的发育成熟以及精子发生可能存在相关关系。 Objective To study the correlation between the expression and activity of hormone-sensitive lipase (HSL) in testis and epididymis and the reproductive maturity of guinea pigs in different developmental stages.Methods The testicular HSL mRNA of guinea pigs in different developmental stages (6-day old,16-day old,and 70-day old) was assessed respectively by RT-PCR.Enzymatic activity was detected by chromatometry at 37℃.The serum testosterone level was measured by radioimmunology.Morphological analysis was performed on HE stained tissue sections of testis and epididymis.Results With the increasing ages of guinea pig,testicular HSL expression and activity raised gradually (P0.05),and the serum testosterone levels increased (P0.01).Histopathological results showed that spermatogenesis emerged in testis and epididymis of 16-day-old guinea pigs and matured in 70-day-old guinea pigs.The HSL mRNA expression and activity were highly correlated with serum testosterone level (r=0.75,P0.01,r=0.96,P0.01).Conclusion The high expression and activity of testicular HSL can be observed in testis and epididymis of mature guinea pigs.There exists the correlation between testicular HSL and testis maturity and spermatogenesis.
