摘要
大豆蛋白序列中隐藏着外啡肽β-酪啡肽-3(β-CM-3)的片段,使得大豆肽食品中含有外啡肽β-CM-3成为可能,因此有必要对大豆肽食品进行外啡肽β-CM-3定性和定量的检测。采用反相液相色谱的方法,以β-酪啡肽-3标准品进行了包括分离柱、流动相的选择、柱温、柱压、流速、检测波长和灵敏度等色谱条件的探索,得出检测条件为波长220nm,柱温25℃,进样量10μL,采用等度洗脱,洗脱条件为:含0·1%TFA的14%乙腈溶液。流速0·9mL/min,洗脱时间为60min。该方法的相对标准偏差为4·07%,平均回收率为95·3%,最小检测浓度为0·4μg/mL。对照标准品保留时间和峰面积,得出酸性蛋白酶/Alcalase=4h/2h酶解液中含有目标外啡肽156·41μg/g。
In the soy protein sequence,there is β-Casomorohin-3(β-CM-3),which may appear in soybean peptide food.Therefore it's necessary to determine the β-CM-3.The separation column,mobile phase,temperature,pressure,wave length and sensitivity were studied based on β-CM-3 standard by reverse phase high performance liquid chromatorgraphy(RP-HPLC).The results showed that the optimum assessing conditions were 220nm of wave length,25℃,10μL ofinlet,isocratic elution,14% methyl cyanide solution including 0.1%TFA,0.9mL/min of flow speed,60min of the elution time.The relatively standard deviation of this method was 4.07%,the average ratio of recovery was 95.3% and the smallest examination density was 0.4μg/mL.And it's determined that there were 156.41μg/g β-Casomorohin-3 in the enzymolysis sample of acidic proteinase/Alcalase=4h/2h.
