摘要
目的研究遗传印记基因PEG10(paternally expressed gene10)在妊娠早期蜕膜组织中的动态表达规律,探讨其在早期胚胎发育及妊娠的维持中的作用。方法收集正常妊娠早期6~11周人工流产蜕膜组织60例,每孕周10例。分别采用半定量逆转录聚合酶反应(RT-PCR)、原位杂交、免疫印迹(Western blot)及激光共聚焦技术检测蜕膜组织PEG10mRNA及蛋白的表达与分布。结果 RT-PCR显示PEG10在各孕周蜕膜组织中均有表达,且表达量随孕周增加逐渐上调,在妊娠第6周表达量较低(0.6743±0.114),随后表达量上升(7周0.7349±0.0083),至第8周达高峰(0.7354±0.0074),继而出现下调趋势(9周0.6340±0.0084、10周0.5901±0.0089、11周0.5261±0.0112)。统计分析,第7、8孕周PEG10表达无显著性差异,其余各孕周两两比较均具有显著性差异(P<0.05);原位杂交、Westernblot及激光共聚焦分析也显示PEG10的表达规律与RT-PCR结果相吻合。结论遗传印记基因PEG10维持一定水平的表达可能对早期胚胎发育和正常妊娠的维持有重要意义。
【Objective】To investigate the role of paternally expressed gene 10(PEG10) in deciduas of normal pregrant women(NP).【Methods】Sixty deciduas tissue from pregnant 6w to 11w were divided into six groups and each groups included ten samples.The expression and distribution of PEG10 in deciduas were examined by RT-PCR,hybridization in situ,western blot and laser scanning confocal miscroscopy.【Results】The PEG10 mRNA and protein were expressed in decidua tissue from pregnant 6w to 11w.Among them,the expression of PEG10 had a gradual increase with the pregnancy weeks added.In RT-PCR,the PEG10 expression was lower at pregnant 6w(0.6743 ± 0.114),from pregnant 7w to 8w,the expression was increased gradually(7w 0.7349 ± 0.0083) and reached the pinnacle in 8w(0.7354 ± 0.0074).And then the pinnacle began to descent(9w 0.6340 ± 0.0084、10w 0.5901 ± 0.0089、11w 0.5261 ± 0.0112).It was a significantly different from pregnant 6w to 11w except 7w and 8w.This expression characteristic was demonstrated by hybridization in situ.? The similar? conclusion was showed ? by Western blot and laser scanning confocal microscopy.【Conclusions】Enough expression of PEG10 may be played an important role in early pregnancy.
