三七总皂苷对肺纤维化小鼠血清中Ⅳ-C型胶原透明质酸的干预作用 被引量:6

摘要 目的:研究三七总皂苷对小鼠肺纤维化血清中Ⅳ-C型胶原及透明质酸的的影响。方法:C57BL/6雄性小鼠60只,分为三七总皂苷高、中、低剂量组,强的松组,模型组,假手术组,每组10只。除假手术组外其余各组小鼠气管内一次性滴注盐酸博莱霉素,假手术组小鼠气管内一次性滴注等体积的生理盐水。造模后第2天开始给药,持续到处死动物的前一天。三七总皂苷高、中、低剂量组分别为120、60、30mg/kg/d,强的松组为0.56mg/kg,假手术组、模型组分别灌服等体积的生理盐水。于28d后处死,观察比较各组小鼠血清中Ⅳ-C型胶原及透明质酸的含量。结果:模型组小鼠肺组织中Ⅳ-C型胶原、透明质酸含量均明显高于假手术组(P<0.01),与模型组相比,三七总皂苷高中低剂量组均能降低小鼠血清中Ⅳ-C型胶原、透明质酸含量,其中中、高剂量组的降低作用较为显著(P<0.01)。结论:三七总皂苷可通过降低肺纤维化小鼠细胞外基质含量而发挥治疗肺纤维化作用。 Objective:To investigate the effect of PNS on pulmonary fibrosis and the hyaluronic acid in mice serum collagen Ⅳ-C.Methods:C57BL/6 mice 60 were divided into PNS high dose group, PNS medium dose group, low dose group, prednisone group, model group, sham operation group, n=10.The mice were given a single intratracheal instillation of bleomycin hydrochloride except mice of sham operation group, while which were given a single intratracheal instillation with saline.The mice were given medicine 2 days after modeling until before they were killed.PNS for high, medium and low dose group were 120, 60, 30 mg/kg/d, respectively; Prednisone was 0.56mg/kg.Sham operation group, model group were given equal volume of saline.At 28d after injection, the serum levels of collagen Ⅳ-Cand hyaluronic acid content were detected.Results:Ⅳ-C collagen, hyaluronic acid in the lungs of mice in model group were significantly higher than that in sham operation group (P0.01), Compared with model group, PNS high dosages could reductionⅣ-C collagen, hyaluronic acid, which in the role of high dose group decreased more significantly (P0.01); Conclusion:PNS could treat pulmonary fibrosis by reducing pulmonary fibrosis in mice.
