摘要
[目的]采用高效液相色谱-蒸发光散射检测法(HPLC-ELSD)建立同时测定芒果中果糖、葡萄糖和蔗糖的方法。[方法]芒果样品处理后,用水提取,用Kromasil NH2柱分离,以乙腈-水(85∶15,V/V)为流动相,使用蒸发光散射检测器进行检测。[结果]果糖、葡萄糖、蔗糖的线性范围分别为0.5~3.0 g/L、0.5~5.0 g/L、0.5~5.0 g/L,在线性范围内各种糖的含量与峰面积有很好的线性关系。样品中糖的加标回收率范围为98.5%~102.8%。用该试验方法测得芒果中果糖、葡萄糖、蔗糖含量分别为2.57%、0.53%、1.86%。[结论]首次建立了同时测定芒果中果糖、葡萄糖、蔗糖的HPLC-ELSD法。该方法具有准确度高、稳定性与重现性好且操作简单等优点,为芒果中糖含量的测定提供了一种准确、灵敏、快速的分析方法。
[Objective] The study aimed to establish the method for synchronously determining the fructose,glucose and sucrose in mango by high performance liquid chromatography with evaporative light scattering detector(HPLC-ELSD).[Method]The treated samples were extracted with water and then separated by Kromasil NH2 column with the acetonitrile and water(85∶15,V/V) as the mobile phase and they were determined by evaporative light scattering detector.[Result] The linear ranges of fructose,glucose and sucrose were 0.5-3.0 g/L,0.5-5.0 g/L and 0.5-5.0 g/L,resp.and the each sugar content had good linear relationship with peak area in the linear range.The average recoveries for all the 5 analytes were in the range 98.5%-102.8%.The contents of fructose,glucose and sucrose in mango determined by this method were 2.57%,0.53%,1.86% resp.[Conclusion] The HPLC-ELSD method for synchronously determining the fructose,glucose and sucrose in mango was established firstly.The method had the advantages of high accuracy,good stability and reproducibility and simple operation and provided an accurate,rapid,sensitive analysis method for determining the sugar content in mango.
