红曲改善大鼠非酒精性脂肪肝胰岛素抵抗的作用机制研究 被引量:7

Effect of Monascus on Improving Insulin Resistance of Rats with Non-alcoholic Fatty Liver Disease and Its Mechanism

摘要 目的研究红曲改善高脂膳食诱导大鼠非酒精性脂肪肝胰岛素抵抗的作用及机制。方法通过高脂饮食建立NAFLD大鼠模型,并利用正常葡萄糖-高胰岛素钳夹技术评估胰岛素抵抗变化,以观察红曲改善胰岛素抵抗的效应。结果实验前(即钳夹开始前60 min)各组大鼠血糖差别较大,模型组大鼠空腹血糖与正常对照组相比有显著升高(P=0.016)。实验开始60 min后(即钳夹开始0 min)各组血糖均达到稳态(5.3 mmol/L)。空腹胰岛素水平高脂饮食组高于正常对照组(P=0.030)。通过钳夹实验发现高脂饮食大鼠的GIR低于普通饮食。结论红曲能降低NAFLD模型大鼠胰岛素抵抗及高胰岛素血症,增加胰岛素敏感性。 Objective To investigate the effect of monascus on improving insulin resistance of rats with non-alcoholic fatty liver disease( NAFLD )induced by high-fat diet, and its possible mechanism. Methods We made the experimental rat model of NAFLD by feeding high-fat diet, and used hyperinsulinemic-euglycemic clamping technique to estimate insulin resistance. Results The difference of blood sugar (BS)in various groups was great before the experiment (60 rain before clamping) . BS in the model group was higher than that in the normal control group(P = 0.016). Sixty minutes after the experiment (0 min of clamping), BS in various groups arrived to homeostasis (5.3 retool/L). Fasting insulin level in high-fat diet group was higher than that in the normal control group (P = 0.030). Results of the hyperinsulinemic-euglycemic clamping experiment showed that the GIR in high-fatty diet group was lower than that in the ordinary diet group. Conclusion Monascus can decrease the insulin resistance and the incidence of hyperinsulinemia, and increase the sensitivity of insulin.
