摘要
目的:探讨罗格列酮对高糖及间歇性高糖诱导大鼠肾小管导管上皮细胞株NRK-52E中NF-κB及肿瘤坏死因子-α(TNF-α)表达的抑制作用。方法:作用72h后,Westernblot检测空白组(0mmol/L葡萄糖)、正常葡萄糖组(5mmol/L葡萄糖)、高糖干预组(25μmol/L葡萄糖)、间歇性高糖干预组(5mmol/L葡萄糖/25mmol/L葡萄糖交替)、罗格列酮干预组(20μmol/L罗格列酮)、罗格列酮高糖干预组(20μmol/L罗格列酮预处理后在高糖培养)、罗格列酮间歇性高糖干预组(20μmol/L罗格列酮预处理后在间歇性高糖培养)中NF-κB及TNF-α及在NRK-52E细胞的表达。细胞ROS含量应用CM2H2DCFDA试剂盒检测。结果:高浓度葡萄糖及间歇性高糖均上调NF-κB及TNF-α表达,ROS含量显著上升,其中在间歇性高糖作用更显著。应用罗格列酮干预后可以下调高糖或间歇性高糖中NF-κB及TNF-α表达,细胞ROS含量下降。结论:罗格列酮可下调高糖及间歇性高糖诱导NRK-52E细胞NF-κB及TNF-α表达。
Objective To explore the inhibiting effect of rosiglitazone on the expressions of NF-κB and TNF-αin NRK-52E cells induced by prolonged or intermittent high glucose level. Methods 72 hours after treatment, the expressions of NF-κB and TNF-α were detected by Western blot in control group (0 mmol/L glucose), normal glucose group (5 mmol/L glucose), high glucose group (25 mmol/L glucose), intermittent high glucose group (between 5 mmol/L and 25 mmol/L glucose), rosiglitazone group (rosiglitazone of 20 μmol/L), rosiglitazone for high glucose group (rosiglitazone of 20 μmol/L and 25 mmol/L glucose), rosiglitazone for intermittent high glucose group (rosiglitazone of 20 μmol/L and 5 mmol/L glucose/25 mmol/L glucose). Intracellular ROS level was detected by CM2H2DCFDA. Results Prolonged or intermittent high glucose level upregulated the expressions of NF-κB and TNF-αand markedly increased ROS level and intermittent high glucose level has a stronger effect. The expressions of NF-κB and TNF-α downregulated and ROS level declined after treatment with rosiglitazone. Conclusions Rosiglitazone can downregulate the expressions of NF-κB and TNF-αin NRK-52E cells induced by high or intermittent high glucose level.
