表没食子儿茶素没食子酸酯对急性酒精中毒小鼠神经行为学改变的影响 被引量:2

Effect of epigallocatechin-3-gallate on neurobehavioral mutation in mice with acute alcoholism

摘要 目的研究表没食子儿茶素没食子酸酯(EGCG)对急性酒精中毒小鼠神经行为学变化及血醇浓度的影响。方法昆明种小鼠随机分为6组,正常组、模型组、EGCG25、75、225mg/kg三个剂量组、阳性药组(葛根胶囊180mg/kg)。给药组灌胃给予相应药物,模型组给予等量生理盐水,0.5h后,按5g/kg灌胃市售56%(v/v)二锅头酒,在0.25、0.5、0.75、1、2h时分别检测各组小鼠神经行为学指标(自主活动、Morris水迷宫、转棒实验)及血醇浓度。结果与模型组比,EGCG75、225mg/kg剂量组在0.25~1h自主活动次数降低,在0.75~2h乙醇小鼠的平衡失调有显著缓解,在0.5~1h血醇浓度降低,EGCG225mg/kg剂量组显著改善乙醇小鼠的空间辨别能力。结论 EGCG可降低血醇浓度,抑制乙醇小鼠中枢神经系统的兴奋性,改善乙醇所致的小鼠空间辨别障碍及共济失调现象。 Objective To investigate the effects of epigallocatechin-3-gallate(EGCG) on the neurobehavioral appearance and blood alcohol concentration of acute alcoholism in mice.Methods Mice were randomly divided into 6 groups:normal group,model group,EGCG(25,75,225 mg/kg) treatment group and Ge Gen capsule(180 mg/kg) treatment group.Mice in the model group were treated with the same saline and mice in the treatment groups were administered with the related drug.After 0.5 h,all groups were infused 56%(v/v)"Er Guo Tou"with 5 g/kg.Then locomotors activity,Morris water maze,balance beam test and blood alcohol concentration were detected at 0.25,0.5,0.75,1 and 2 h after infused alcohol.Results Compared with model group,EGCG(75,225 mg/kg) had significantly decreased the locomotors activities from 0.25 to 1 h,moderated ataxia from 0.75 to 2 h and decreased blood alcohol concentration from 0.5 to 1 h.Especially,EGCG(225 mg/kg)had significantly improved spatial discrimination disorders.Conclusion EGCG can decrease blood alcohol concentration,suppress the irritability of central nerve system and improve spatial discrimination disorders and ataxia.
