摘要
以虫草素含量为指标,采用正交实验优化了适合拟黑虫草(Cordyceps nigrella)菌株cnig-56固体发酵的培养基,并探讨了该菌株固体发酵的最佳条件。结果表明,菌龄为3d的拟黑虫草菌种按10%接种量接入优化的固体培养基上(配方为大米粉9.9%,青稞粉6.6%,玉米粉9.9%,蚕蛹粉3.3%,粗麸皮70.15%,KH2PO40.05%,MgSO40.1%)于24℃发酵培养30d,其产物的虫草素含量最高为101μg/g。
Orthogonal testing was used to optimize selected parameters affecting cordycepin production by Cordyceps nigrella (strain cnig-56) grown under solid-state conditions. Highest cordycepin levels (101 μg/g) were recorded after 30 d cultivation at 24 ℃ on a medium consisting of 9.9% rice powder,6.6% highland barley powder,9.9% corn powder,3.3% dried silkworm chrysalis powder,70.15% unrefined wheat bran,0.05% KH2PO4,0.1% MgSO4·7H2O,a solid:water ratio of 1:1.2,and using 3-day-old mycelium (10% w/w) as inoculum.
