中国梨醇-酰基转移酶基因的克隆及遗传多态性 被引量:3

Alcohol-acyltransferase (AAT) gene cloning and genetics polymorphism of six Chinese pear cultivars

摘要 以6个中国梨品种为实验材料,对基因组DNA进行醇-酰基转移酶(AAT)基因的特异性PCR扩增、克隆、DNA序列测定和生物信息学分析,研究了中国梨AAT基因的遗传多态性。结果表明,浓香、微香、无香梨品种中均克隆到约1 600 bp大小的1-4条AAT基因。其中外显子部分存在1个HXXXD活性区和4个可变区,内含子中至少存在8处长度缺失和突变。基因结构分析表明,这些AAT基因同属于家族cl03601的转移酶基因家族pfam02548。系统发育分析表明,本实验所克隆到的中国梨AAT基因关系最近,与网上已知序列的苹果和西洋梨的关系略远。 Six Chinese pear cultivars were used as materials for PCR amplifying and cloning AAT-gene.In addition,sequence bioinformatics analysis were performed.The results show that there were AAT genes in ten Chinese pear cultivars with heavy-aroma,light-aroma,no-aroma,furthermore,1-4 AAT gene with the similar size were cloned,which exons including an active site motif HXXXD and one hyper variable region,and there were eight length deletions and mutation s at least in the intron.AAT-gene structure were analyzed which were cloned.The results show that they belong to transferase gene family pfam02548 of family cl03601.The phylogenetic analysis shows that the AAT gene cloned in this experiment is clustered into one class,the relationship among the Chinese pear varieties was closer,and online known sequence relationship between the apples and pears was a little farther.
