摘要 对牛分支杆菌三价DNA疫苗工程菌进行了高密度培养的探索。通过摇瓶培养试验对3种培养基进行了筛选,选择出具有高密度培养潜力的半合成培养基作为中试规模的发酵罐培养基;经对培养温度、溶氧、pH、转速、补料方式等各种条件优化,培养的工程菌密度OD600达到22.92,细菌湿重达到27.173 g/L,质粒产量为1.44 mg/g湿菌。试验还对培养物质粒的稳定性进行了证实。 Engineering bacteria with high density cultivation in Mycobacterium bovis DNA Vaccine were explored in this article.Three kinds of media were screened by the flask experiments,and a semi-synthetic medium which has the potential of high-density fermentation as the pilot scale medium was selected in the fermentor.The culturation temperature,dissolved oxygen,pH,rotational velocity,and feeding methods etc.were optimized,the E.coli density of fermentation is up to OD600 22.92 with the semi-synthetic medium,and the humid weight of engineering bacteria is 27.173 g/L.The outcome of plasmid is 1.44 mg/g.The stability of the plasmid in the engineering bacteria was also verification.
