摘要
目的探讨人参皂苷Rg1对脑缺血-再灌注大鼠海马CA1区神经元的保护作用及机制。方法将健康雄性SD大鼠随机分为假手术组,缺血再灌注组,人参皂苷Rg110、20、40mg/kg组,尼莫地平阳性药物对照组。采用Nissl染色检测大鼠缺血再灌注后海马神经元损伤,并采用免疫印迹方法检测p-JNK、p-c-jun、Bax的表达情况。结果假手术组海马神经元数量正常,p-JNK、p-c-jun、Bax含量最低;与假手术组比较,缺血再灌注组神经元数量显著降低,p-JNK、p-c-jun、Bax含量显著增高;各给药组与缺血再灌注组比较,海马神经元损伤减轻,p-JNK、p-c-jun、Bax含量降低,其中人参皂苷Rg140mg/kg组效果更明显。结论人参皂苷Rg1对大鼠脑缺血-再灌注损伤具有保护作用,可能与抑制脑组织p-JNK、p-c-jun、Bax的表达有关。
Objective To explore the protective role and mechanism of Ginsenoside Rg1 on the ischemic-reperfusion injuried neurons in hippocampus CA1 of rat.Methods Healthy male SD rats were randomly divided into the sham-operative group,focal cerebral ischemia-reperfusion group,Ginsenoside Rg1 treated groups(Rg1 10,20,40 mg/kg),and Nimodiping treated group.Nissl staining and Western blot were used to detect neurons and the expression of p-JNK,p-c-jun and Bax.Results The percentage of neurons was decreased and the expressions of p-JNK,p-c-jun and Bax in hippocampus CA1 zone were increased significantly in ischemia-reperfusion group than in sham control group.However,the percentage of neurons was increased and the expressions of p-JNK,p-c-jun and bax in hippocampus CA1 zone were decreased in Rg1 treated group than in ischemia-reperfusion group(P〈0.05),paticularly in 40 mg/kg subgroup.Conclusion The protective effect of Ginsenoside Rg1 on the ischemia-reperfusion injuried neurons might be related to the inhibition of expressions of p-JNK,p-c-jun and Bax in hippocampus CA1 zone.
