摘要
崇明岛东滩湿地是我国主要的湿地之一.试验对东滩湿地的光滩、海三棱藨草根部土壤和芦苇根部土壤样品用稀释的R2A培养基分别进行培养.通过平板计数得到每克光滩样品、海三棱藨草样品、芦苇样品中的细菌数量分别为8.6×106cfu,1.3×107cfu,1.05×108cfu.对所分离到的84株菌株的16S rRNA基因序列的多样性指数进行分析,结果表明光滩样品的多样性最高.通过与公共数据库中标准菌株序列的比较以及系统发育树分析确定了这些菌株的分类地位,结果表明分离到的菌株分布于γ-变形菌门、拟杆菌门、厚壁菌门、α-变形菌门和放线菌门的30个属,在光滩中分离到8个其他样品中没有的属.
Chongming Dongtan is one of the main wetlands in China. Here, three soil samples from tidal flat,Scirpus triqueter root region and Phragmites australis root region were cultivated by diluted R2A medium. Through plate counting method, the bacterial numbers in one gram soil were 8. 6×10^6 cfu in tidal flat sample, 1.3×10^7 cfu in Scirpus triqueter sample and 1.05×10^8 cfu in Phragrnites australis sample. According to the results of 16S rRNA gene sequences of isolated 84 strains, the diversity index of the tidal flat was the highest. Comparing with 16S rRNA gene sequences of standard strains and the constructed phylogenetic tree, the taxonomy of these strains was defined. These isolates belong to 30 genera of γ-Proteobacteria, Bacteroidetes, Firmicutes, a-Proteobacteria, and Actinobacteria. And up to 8 genera related bacteria were only isolated from tidal flat sample.
