摘要
采用胶原酶的单酶冷消化法,分离培养37例肾细胞癌患者的肿瘤浸润淋巴细胞(tumor infiltra-ting lymphocytes,TILs)。结果发现,采用该法分离培养的 TILs 呈现以激活的 T 淋巴细胞为主、高增殖活性和高杀伤活性的特征。逆转录-多聚酶链式反应(RT-PCR)检测结果提示,TILs 激活扩增后主要分泌产生肿瘤坏死因子-α(TNF-α),其可能在 TILs 发挥抗肿瘤活性中起重要作用。应用结果显示,治疗后患者外周血淋巴细胞表型分析,90%以上患者 CD3^+细胞和70%患者 CD8^+细胞得到提高。机体的细胞免疫水平提高。
We isolated tumor infiltrating lymphocytes (TILs) from 37 renal cell cracinoma tissues digested with collagenase Ⅱ at 4℃.The cells have some notable features,mainly composed of activated T lymphocytes,high reproduction rate and powerful antitumor activity.The detection results with RT-PCR of cytokines showed that TNF-a secreted by TILs might be related to the antitumor mechanism of TILs,and some cytokines secreted by tumor cells might play an important role in their occurrence and development.We can also conclude from the results of study on the clinical application of TILs that the cellular immunity was improved in patients accepting the therapy.
