摘要
清远麻鸡属于小型优质鸡种,为探讨其优良肉质形成的分子机理,此研究以隐性白羽鸡作为对照,以CAPN1基因作为影响鸡肌肉嫩度性状的候选基因,采用PCR-SSCP方法对清远麻鸡和隐性白羽鸡的CAP]N1基因外显子4部分序列、内含子4和外显子5部分序列进行单核苷酸多态性分析,共检测到AA、AB、AC、BB、BC和CC 6种基因型。测序结果发现,AA型个体序列与Genbank上登录的序列一致;BB型个体在2418处存在5 bp的缺失突变,在2446处存在T/C突变,在2484处存在C/T突变,在2508处存在C/T突变;CC型个体在2418处存在5 bp的缺失突变,在2446处存在T/C突变,在2484处存在C/T突变。统计了不同基因型在不同品种鸡中的频率分布,结果表明基因型频率和基因频率在2个鸡种中的分布存在显著差异。
Qingyuan partridge chicken is a light-body type breed with good meat quality,in order to explore the molecular mechanisms of its good meat quality,we chose CAPN1 gene as candidate gene for chicken meat tenderness,and Recessive White chickens as comparison in the present study.PCR-SSCP and sequencing methods were used to investigate single nucleotide polymorphisms in CAPN1 gene.Six genotypes were detected,i.e.AA,AB,AC,BB,BC and CC.Sequencing results revealed that AA genotype was coincidence with the sequence accessed in Genbank,there is a 5 bp deletion mutation at position 2418,a T/C point mutation at position 2446,a C/T mutation at position 2484,and a C/T mutation at position 2508 in BB-type individuals;there is a 5 bp deletion mutation at position 2418,a T/C point mutation at position 2446,and a C/T mutation at position 2484 in CC-type individuals.Allele frequencies and genotype frequencies distribution were significant different in the two chickens.
