摘要
目的:观察保肺定喘汤对肺心病的防治作用并探讨其作用机制。方法:60只SD大鼠随机分为正常对照组,模型组,保肺定喘汤高、中、低剂量组和硝苯地平组。采用野百合碱(60mg/kg)腹腔注射建立肺心病大鼠模型,连续用药28天后处死大鼠。放免法测定血中TXB2、6-keto-PGF1α、ET-1水平,酸还原法测定NO含量;利用RT-PCR技术测定肺组织ET-1mRNA的变化,称取RV和(LV+S)重量,计算右心肥厚指数〔RV/(LV+S)〕;计算机图象分析方法观察肺小动脉病理改变。结果:模型组大鼠右心肥厚指数明显增加,血中TXB2、ET-1及肺组织ET-1mRNA水平升高,6-keto-PGF1α和NO含量降低,肺小动脉管壁变厚、管腔变小(P均<0.01)。各治疗组均能降低右心肥厚指数、血中TXB2、ET-1水平及肺组织ET-1mRNA表达,升高血中6-keto-PGF1α和NO含量,肺小动脉管壁有不同程度变薄,管腔变大(P<0.01~0.05)。结论:保肺定喘汤可通过调节TXB2、6-keto-PGF1α、NO、ET-1的代谢和减轻肺血管重构,在一定程度上防治肺心病。
Objective: To observe the effects of baofei dingchuan decoction on TXB2, 6-keto-PGF1α,NO and ET-1 levels in rats with pulmonary heart disease.Methods: 60 rats were randomly divided into 6 groups, normal, model,nifedipine, baofei dingchuan decoction low, middle and high dose groups. Models with pulmonary heart disease were made by interperitoneal monocrotaline(60mg/kg) once, each group was given corresponing management(normal and model groups were given 0.9%NaCl,nifedipine group was given nifedipin 10mg/kg,baofei dingchuang decoction low,middle,high dose groups were given baofei dingchuan deoction 3.1,6.2,12.4g/kg)for 28 days, the plasm TXB2, 6-keto-PGF1α,NO and ET-1 levels were determined by radioimmunoassay and ET-1 mRNA expression of lung was examined by RT-PCR.The right ventricular hypertrophy index was calculated, arteriole pathologic changes of lungs were observed by light microscope and image analysis system.Results: The right ventricular hypertrophy index of the model group rats was obviously increased, the plasm TXB2,ET-1 levels and ET-1 mRNA expression of lung were elevated significantly, 6-keto-PGF1α and NO contents were lowered, the pulmonary vascular wall got thicker and lumens got more narrow. All these changes of model group were improved in each treated group at different degree, the right ventricular hypertrophy index was decreased, the plasm TXB2,ET-1 levels and ET-1 mRNA expression of lung were reduced, 6-keto-PGF1α and NO contents were increased, the pulmonary vascular wall got thinner and lumens got bigger,there were singnificant difference between each treated group and model grou(P<0.05~0.01).Conclusion: Baofei dingchuan decoction can prevent and treat pulmonary heart disease by regulating TXB2, 6-keto-PGF1α,NO and ET-1 levels and abating pulmonary arteriole pathologic changes.
