摘要
以‘西豆3号’等3个大豆品种为材料,以胚芽尖为外植体,研究种子萌发时间、植物生长调节剂对不定芽分化的影响以及生长调节剂对不定芽生根的影响,建立高效再生体系.结果表明种子在1/2MS培养基上萌发24h后,将胚芽尖接种在BM+5mg/LBA培养基上培养24h,然后转入BM+0.4mg/LBA+0.4mg/LIBA培养基诱导不定芽,出芽率达64.91%~68.89%;将长3~5cm的不定芽在BM+0.5mg/LBA+1.5mg/LIBA培养基中培养7d后,转入BM基本培养基中生根,生根率达22.22%~26.32%.
. In order to establish an efficient embryonic tip regeneration system for soybean (Glycine max Merr. ), the embryonic tips of three soybean cultivars, including Xidou No 3, were used as explants to study the effects of seed germination time and growth regulator concentration on shoot differentiation and plantlet rooting of this regeneration system. Mature soybean seeds were made to germinate on 1/2 MS medium for 24 h, arid their embryonic tips were collected and cultured on BM medium (basic medium) sup- plied with 5 mg/L BA for 24 h and then transferred to BM medium supplied with 0.4 mg/L BA and 0. 2 mg/L IBA. A shoot regeneration percentage of 64.91 %-68.89 % was achieved. Adventitious shoots 3- 5cm long were later cultured on BM medium supplied with 0.5 mg/L BA and 1.5 mg/L IBA for 7 days and then transferred to BM medium without any hormone. Finally, 22.22 %-26.32 % of them rooted readily.
