摘要
RNA干扰(RNAi)是一种在动植物中广泛存在的、由双链RNA诱发的、同源mRNA特异性沉默的过程,包括小干扰RNA生成的起始阶段及靶mRNA降解的效应阶段。已成为分子生物学研究中最为活跃的热点之一;在肿瘤研究中,RNAi在抑制肿瘤抗凋亡基因、沉默癌基因、调控细胞周期、抗血管生成、增强放化疗敏感性及基因功能研究等有较广泛的应用。
RNA interference(RNAi) ,which is a phenomenon widely existing in animals and plants,is a mRNA-specific silence process, induced by homologous double-stranded RNA, including initial stage( generation of small interference RNA, siRNA) and effect stage ( degradation of target mRNA). RNAi has been one of the most active hot spot in molecular biology research. In the research of cancer, RNAi has been broadly applied in inhibiting cancer anti-apoptosis gene, silencing oncogenes, cell cycle regulation, anti-angiogenesis, enhancing chemotherapy sensitivity and gene function studies.
