摘要
目的:从蚯蚓粪和黄粉虫沙中筛选对该土传病害病原菌——尖孢镰刀菌(Fusarium oxysporum)具有拮抗效果的微生物菌株,可以有效控制黄瓜枯萎病(CucumberFusarium wilt)的发生。方法:经PDA平板对峙实验、紫外线诱变处理和黄瓜种子胚轴抑制试验,得到具有拮抗效果的菌株,通过真菌的18S rRNA的PCR扩增及克隆、18S rRNA的全序列分析等手段。结果:从23株具有拮抗效果的菌株中得到突变菌株syx-2及该菌株的18S rRNA基因序列。结论:syx-2为白地霉(Geotrichum candidum)的一个变种,对黄瓜枯萎病活体拮抗作用可高达81.1%,具有明显的抑制作用。
Objective:Some strains which had antagonism effect on pathogen-Fusarium oxysporum were screened from wormcast and mealworm faeces.Method: By PDA agar plates,ultraviolet radiation and cucumber radicel inhibition test,some strains had got which had efficient antagonism.Then,the gene sequence of the 18S rRNA of the strain fungi by PCR amplification and clone of the 18S rRNA were studied.Result: The mutant strain syx-2(FJ754318.1) with high antagonism effect and its sequence analysis of 18S rRNA had got.Conclusion: This fungi was a variety of Geotrichum candidum,its antagonism effect on cucumber Fusarium wilt could reach 81.1%.
