Relationship between Alzheimer disease and brain-derived neurotrophic factor

摘要 目的探讨阿尔茨海默病(Alzheimer disease,AD)患者脑脊液脑源性神经营养因子(Brain-derived neurotrophic factor,BDNF)的变化,以及外源性BDNF的治疗效果。方法选取2006年1月-2008年5月在我院住院的24名AD患者和26例非痴呆脑梗死患者,采用酶联免疫吸附法检测他们脑脊液脑源性神经营养因子的水平,并对其中的12名AD患者经椎管注入外源性BDNF进行治疗,以治疗前后简易智能精神状态量表(MMSE)评分差值来评估疗效。结果AD患者脑脊液BDNF浓度显著低于非痴呆脑梗死患者(t=-4.927,P=0.000),并且外源性BDNF能提高MMSE的评分(F=56.067,P=0.000),且不受年龄的影响(F=1.667,P=0.211)。结论脑脊液BDNF可作为AD的诊断标志物,并且BDNF能很好地改善AD患者的认知能力而不受年龄的影响。 Objective To study the variation of brain - derived neurotrophic factor (BDNF) from cerebro- spinal fluid and the therapeutic efficacy of exogenous BDNF on Alzheimer disease(AD). Methods The inpatients of 24 AD and 26 cerebral infarction of undementia were chosen from January 2006 to May 2008, and the BDNF from cerebrospinal fluid were detected by enzyme - linked immunosorbant assay. The therapeutic efficacy of 12 AD patients injected exogenous BDNF via vertebral canal was evaluated with the difference of Mini - Mental State Exami nation (MMSE) before and after treatment. Results The concentration of BDNF from cerebrospinal fluid at AD patients was significantly lower than that of cerebral infarction of undementia( t = - 4. 927 ,P = 0. 000), and the exogenous BDNF was able to elevate the AD scores of MMSE (F = 56. 067 ,P = 0.000) without the influence of age (F = 1. 667,P = 0.211 ). Conclution The BDNF from cerebrospinal fluid can be a diagnosis maker of AD, and the perceptibility of AD patients was improved very well by the BDNF without the influence of age.
