雷公藤内酯醇降低T淋巴细胞核因子-κB的活性 被引量:82

摘要 目的:研究雷公藤内酯醇对T淋巴细胞核因子-κB(NF-κB)活力的影响,进一步研究雷公藤内酯醇免疫抑制作用的机制。方法:利用凝胶迁移率实验(EMSA)检测不同浓度的雷公藤内酯醇对于普通培养状态下和同时使用佛波脂/植物血凝素(PMA/PHA)激活的Jurkat细胞中NF-κB活力的影响。结果:在普通培养状态下和同时使用PMA/PHA激活的Jurkat细胞中均存在一定活力的NF-κB,使用PMA/PHA处理可使Jurkat细胞中NF-κB的活力显著的升高,雷公藤内酯醇可以降低两种培养状况下的Jurkat细胞中NF-κB活力,并以激活状态下更为显著。结论:雷公藤内酯醇降低T淋巴细胞中NF-κB活力的作用可能是其免疫抑制效应的分子机制之一。 Abstract OBJECTIVE Triptolide is one of the most potent ingredients for the immunosuppressive effect of Tripterygium Wilfordii Hook f.To further explore the mechanism of triptolide,the inhibitory effect of triptolide on the activity of nuclear factor κB(NF κB),an important transcription factor,was investigated in the present study. METHODOLOGY Human T lymphocyte cell line Jurkat cells were used in the study.Jurkat cells were divided into two groups,one was stimulated with PMA/PHA(25μg/L and 2μg/L),the other was not stimulated.Both groups were treated with or without triptolide at different concentrations(2μg/L or 10μg/L)for 1 hour.The activity of NF κB was measured by electrophoretic mobility shift assay. RESULTS PMA/PHA rapidly stimulated the activation of NF κB in Jurkat cells.Triptolide decreased the activity of NF κB in both groups,in a dose dependent manner.This effect was more obvious in the cells stimulated by PMA/PHA. CONCLUSION These results demonstrate that triptolide is a potent inhibitor of NF κB activation in T lymphocyte.This effect may explain some of the known immunosuppressive properties of triptolide.
