摘要
目的制备抗AIF多克隆抗体。方法用人工合成的AIF抗原肽免疫实验动物制备多克隆抗体并纯化。利用ELISA和Western blot方法检测多克隆抗体的效价和特异性。结果实验获得的血清抗AIF抗体经间接ELISA法检测,效价达到1∶128 000。Western blot检测表明抗体效价高、特异性强。结论实验获得了高效价的特异性的抗AIF抗体,为进一步研究肿瘤的抗凋亡机制提供新的方法。
Objective To prepare AIF polyclonal antibody.Method Rabbit was immunized with AIF protein,purified polyclonal antiserum was analyzed by ELISA and Westem blot.Result The detection of indirect ELISA method showed that titer of serum AIF antibody reached 1 ∶128 000.Western Blot method showd that the antibody has high specificity.Conclusion This study obtains high titer specific AIF antibody,which provides theoretical basis for anti-apoptosis mechanism of human esophageal carcinoma.
