摘要
目的以废弃蛋膜为原料,用碱性蛋白酶水解制备小分子多肽。方法在单因素实验的基础上以水解度、肽质量分数为响应值,采用4因素(时间、温度、酶用量、pH)3水平的响应面(Re-sponse SurfaceMethod,RSM)分析法根据回归分析确定影响因子。结果在分析各因素的显著性和交互作用后,得到最佳水解条件:时间2.3h、温度48℃、酶用量2.0%、pH9.1,蛋膜水解度(DH)达到18.23%,肽含量达到62.38%。结论为废弃蛋膜的综合利用提供了一条思路。
Aim In order to obtain the bioactive peptides, the discarded eggshell membrane hydrolyzed by alkaline protease. Methods On the basis of single factor test, four factors (time, temperature, pH value, enzymatic dosage) and two levels' test by response surface methodology (RSM) were designed from DH, peptide content as response values. Results Through the test of response surface, it was found that the optimum conditions were : time 2. 3 h, temperature 48℃, enzymatic dosage 2. 0% ,pH 9. 1. Under these conditions, DH was 18.23 %, and the peptide content was 62. 38%. Conclusion A way was provided to utilize the discarded eggshell membrane.
