ERCC1基因表达与磁性Fe_3O_4纳米颗粒逆转卵巢癌细胞耐药性的关系 被引量:3

The relationship between the expression of ERCC1 genes and Fe_3O_4-magnetic nanoparticals with drug resistance in ovarian carcinoma cells

摘要 目的探讨磁性Fe3O4纳米颗粒(Fe3O4-MNPs)逆转人卵巢癌顺铂耐药细胞株(SKOV3/DDP)的耐药性的可能性及其机理。方法将SKOV3/DDP分为对照组、顺铂组、Fe3O4-MNPs组、顺铂+Fe3O4-MNPs组等4个组,流式细胞技术测定细胞凋亡、电感耦合等离子体原子发射光谱法(ICP-AES)测细胞内顺铂的含量,及RT-PCR检测切除DNA修复基因ERCC1 mRNA在SKOV3/DDP中的表达。结果在顺铂+Fe3O4-MNPs组中,细胞凋亡率及细胞内铂含量均高于顺铂组(P<0.05);ERCC1 mRNA的表达明显低于顺铂组(P<0.05)。结论Fe3O4-MNPs可以逆转卵巢癌细胞对顺铂的耐药性,其作用可能是通过提高细胞内铂浓度、下调ERCC1表达来逆转SKOV3/DDP对顺铂的耐药。 Purpose To determine whether Fe3O4-MNPs can reverse the cisplatin (DDP) resistance to ovarian carcinoma cells (SKOV3/DDP)and to explore its mechanisms.Methods The SKOV3/DDP strains were divided into DDP treatment ( DDP group ), Fe3O4- magnetic Nanopartieals treatment ( Fe3O4- MNPs group ), and DDP + Fe3O4-MNPs treatment(DDP + Fe3O4-MNPs group)and control group. The apoptotic ratios were detected by flows cytometry (FCM), and the intracellular platinum levels in different groups were detected by ICP and the expressions of excision repair cross complement-1 (ERCC1) were detected by reverse transcription polymerase chain reaction (RT-PCR) in different groups. Results In the DDP + Fe3O4-MNPs groups,the apoptotic ratios and the intracellular platinum levels were higher. The expression of ERCC1 was decreased compaired with other groups. Conclusion Fe3O4-MNPs can reverse the DDP resistance to the ovarian carcinoma cells (SKOV3/DDP) ,and the effect may be ascrbed to increase the intracellular platinum levels and decrease the expressions of ERCC1 gene.
