摘要
目的:提取分离茜草(RubiacordifoliaL.)中的活性糖蛋白,并进行结构分析。方法:用DEAE-纤维素和SephadexG-150柱层析纯化茜草粗多糖,得到糖蛋白QC。用高效液相色谱法(HPLC)(IonpakS-805柱)检查纯度并测定相对分子质量。用薄层层析色谱法(TLC)、气相色谱法(GC)、气-质联用色谱法(GC-MS)及红外光谱(IR)分析其多糖的结构。结果:糖蛋白QC经HPLC检查为单一对称峰,说明其为均一糖蛋白,QC的平均相对分子质量为401689;其蛋白含量为7.6%。TLC和GC分析表明,QC由Rha、Ara、Xyl、Man、Glc、Gal及半乳糖醛酸组成,其中性单糖摩尔比为1.00∶1.29∶1.86∶0.99∶2.97∶3.04。IR分析表明,其含β型糖甙键。GC-MS分析表明,Gal、Glc各6种连接方式,Man、Xyl、Ara、Rha分别有3、1、2、2种连接方式。各糖基的连接顺序有待于进一步的研究。结论:茜草中分离得到的糖蛋白QC是一种由多种糖残基组成的多分支的杂多糖蛋白,初步药理实验表明其具有明显的清除自由基的作用,清除率为93.24%。
Objective: To isolate the bioactive glycoprotein from Rubia cordifolia L. and to elucidate its structure. Methods: Glycoprotein QC was isolated and purified by decocting with water,alcohol precipitation, ion exchange (DEAE cellulose) and gel filtration (Sephadex G 150) chromatography. Its homogeneity was proved by HPLC on Ionpak S 805 column. Its structure was analyzed by TLC, GC, GC MS and IR spectrometry. Results: The average MW of QC was 401 698 . According to TLC and GC analysis, the monosaccharide residues of QC were rhamnose, arabinose, xylose, mannose, glucose, galactose and galacturonic acid. The molar ratio of neutral sugars was 1.00∶1.29∶1.86∶0.99∶2.97∶3.04. GC MS Analysis showed that QC was highly branched in structure. IR Indicated that it contains β glucoside bond. Conclusion: QC Is a complex branched glycoprotein composed of several kinds of monosaccharide residues from Rubia cordifolia L. Preliminary pharmacological tests showed that QC had strong scavenging effects on free radicals with a scavenging rate of 93.24%.
