PRRSV SX-1株的分离与NSP2和ORF5基因的克隆及变异分析 被引量:5

Isolation of Porcine Reproductive and Respiratory Syndrome Rirus and Mutation Analysis of Its Nsp2 and ORF5 Gene

摘要 从山东莘县恒顺猪场发病猪群中分离到PRRSV病毒(SX-1株)。用RT-PCR法对该分离株的NSP2和ORF5基因进行了扩增和克隆,并对其进行核苷酸序列测定和分析。结果表明,PRRSV SX-1株的NSP2发生30个氨基酸的不连续缺失,缺失位置与JXA1毒株相同;ORF5基因与JXA1、CH-1a、VR2332核苷酸同源性分别为98.8%、94.9%、88.9%,氨基酸同源性分别为99.0%、93%、87.5%。基因系统发育树分析结果显示,SX-1与JXA1、HUN株的亲缘关系很近,与CH-1a亲缘关系较近,与VR2332、CC-1、RespPRRS/ReproMLV等毒株处于不同分支。 A strain of porcine reproductive and respiratory syndrome virus(PRRSV)was isolated from Hengshun pig herd of Xin county with high fever syndrome in Shandong and it was designated as SX-1 strain. The NSP2 gene and ORF5 gene of SX-1 strain were amplified by RT-PCR and sequenced. The results showed that SX-1 strain was classified into the North American and it had discontinuous 30 amino acids deletions in NSP2. The ORF5 of SX-1 strain shared 98.8% ,94.9% and 88. 9% of nueleotide homology , and 99% ,93%,64.3% of amino acid identity , with those of JXA1, CH-1a and VR2332. The phylogenetic tree revealed SX-1 had closer relationship with JXA1 and HUN strains than that of CH-1a strain.
