摘要
目的观察大鼠颈总动脉球囊损伤后血凝素样氧化低密度脂蛋白受体-1(lectin like oxidized low density lipoprotein receptor-1,LOX-1)的表达及西洛他唑对其表达的影响,探索西洛他唑抑制内膜增生的机制。方法24只健康雄性SD大鼠按完全随机分组法分成假手术组、模型组、西洛他唑组(n=8)。观察球囊损伤后内膜增生情况并计算内膜、中膜面积比值,检测血清MDA含量,RT-PCR法和免疫组化法分别检测各组球囊损伤后大鼠颈总动脉血管壁组织中LOX-1mRNA和蛋白的表达。结果西洛他唑组内膜/中膜面积比值(IA/MA)、LOX-1mRNA及蛋白表达水平、血清MDA含量显著低于模型组,差异有统计学意义(P<0.05)。结论西洛他唑可显著抑制内膜增生,抑制LOX-1表达及其介导的氧化应激,这可能是西洛他唑抑制内膜增生的机制之一。
Objective To explore the expression of leetin-like oxidized low-density lipoprotein receptor-1 ( LOX-1 ) in a rat model after balloon arterial injury and investigate the effect of cilostazol on the mRNA and protein expressions of LOX-1. Methods Twenty-four male SPF SD rats were randomly divided into 3 groups (n = 8 for each group) , sham operation group, balloon arterial injury group and eilostazol treatment group. For the rats from balloon arterial injury group and cilostazol treatment group, the left common carotid artery were injuried by a balloon catheter. The expression of LOX-1 mRNA and protein in neointimal hyperplasia after balloon arterial injury were examined by RT-PCR and immunohistochemistry respectively. The contents of MDA in blood serum was examined by TBA. Results LOX-1 mRNA and protein were expressed in the neointimal hyperplasia after balloon arterial injury but not in the sham-operation group. The expression level of LOX-1 was significantly lower in the eilostazol treatment group than that in the model group (P〈0.05 ). The content of MDA in the blood was also significantly lower in the cilostazol treatment group than in model group (P〈0.05). Conclusion LOX-1 is highly expressed in neointimal hyperplasia after balloon arterial injury and cilostazol significantly inhibits the neointimal hyperplasia by suppressing the expression level of LOX-1 and the contents of MDA in blood serum.
