摘要
桑叶多酚有消除机体内自由基毒性的作用,为桑叶中具有药用和保健功能开发价值的成分之一。运用二次回归正交旋转组合设计法,研究了桑叶多酚提取工艺中原料溶剂乙醇的浓度,以及原料的质量浓度、原料溶液pH值、提取次数等因素对桑叶多酚得率的影响,建立了具有良好预测性能的提取条件数学模型,确定了最优提取工艺条件。试验结果表明,在桑叶多酚提取工艺条件各因素中,对桑叶多酚得率影响作用大小的顺序为原料溶液pH值>原料的质量浓度>溶剂乙醇的浓度。最优提取工艺条件为:以70%乙醇作原料溶剂,原料质量浓度0.025g/mL,原料溶液pH值4。在此优化工艺条件下,从桑叶中提取桑叶多酚的质量比为16.11 mg/g。
Being capable of eliminating toxicity of free radicals in organisms,the polyphenols from mulberry leaves are one of the valuable components which can be developed for use of medicinal and health-care functions.By means of quadratic regression rotational combinational design,the effect of ethanol concentration,material mass concentration,pH of material solution and extraction times on the yield of polyphenol from mulberry leaves was studied.A mathematical model of extraction conditions with good prediction performance was established and an optimum condition for polyphenol extraction was obtained.The result showed that the ranking of the factor was pH value〉mass concentration〉ethanol concentration,according to their influence on the yield of polyphenol.The optimum condition for the extraction was as following: 70% ethanol as extraction solvent,material mass concentration 0.025 g/mL,and pH 4.Under such conditions,the yield of polyphenol from mulberry leaves was 16.11 mg/g.
