摘要
目的观察血小板衍生生长因子(PDGF)对肝星状细胞c—jun基因表达的影响。方法在培养的肝星状细胞株中加入不同浓度的PDGF(终质量浓度8、40、200ug/L),于8、24、48、72h4个时间点分别收集细胞,提取肝星状细胞总RNA;用逆转录一聚合酶链反应(RT-PCR)方法测定c-jun的基因表达水平。结果PDGF3组HSC—T细胞c-jun基因表达水平在8h(1.71±0.20、2.30±0.26、3.30±0.29)、24h(1.46±0.13、1.98±0.19、2.73±0.23)、48h(1.11±0.10、1.63±0.16、2.32±0.21)、72h(0.77±0.09、1.23±0.10、1.91±0.16)4个时间点均显著高于对照组;随剂量加大,HSC-T6细胞c-jun基因表达水平逐渐升高;3组间差异有统计学意义(P〈0.01)。结论PDGF可显著增强肝星状细胞c—jun的基因表达。
Objective To investigate the effects of platelet growth factor (PDGF) on c-jun gene expression in hepatic stellate cells. Methods Hepatic stellate cells (HSC-T6) were cultured in medium containing different concentrations of PDGF ( 8,40,200ug/L respectively), and HSCs were harvested at different incubation periods (8,24,48,72 h). Total RNA of HSCs was isolated ,and the c-jun gene expression level was detected by reverse transcription polymerase chain reaction. Results The c-jun gene expression level of HSCs in low (8 ug/L) ,middle (40ug/L) and high (200 tLg/L) PDGF groups was significantly higher than in control group after exposure to PDGF at 8 h ( 1.71 ± 0.20,2.30 ± 0.26,3.30 ± 0.29) ,2d h (1.46 ±0.13,1.98 ±0.19,2.73 ±0.23) ,48 h (1.11 ±0.10,1.63±0.16,2.32±0.21) and 72 h (0.77 ± 0.09,1.23 ± 0. 10, 1.91 ± 0. 16), respectively. The c-jun gene expression levels in HSCs were enhanced gradually with the increased dosage of PDGF. There was significant difference in the c-jun gene expression among three PDGF groups. Conclusion PDGF may strongly up-regulate the c-jun gene expression in HSCs.
